Spheroids & Live/Dead Detection
Some spheroids permit the visualization of individual cells, enabling single-cell live/dead quantification on a per-spheroid basis.
The ratio of live to dead cells can be obtained directly within the WiSoft Athena software environment and can be normalized to total cell count or live vs. dead.
Spheroid Solidity (S)
Solidity, S, is the measurement of the overall concavity of a particle. As the particle becomes more solid the image area and convex hull area approach each other resulting in a solidity value of one.
However, as the particle form digresses from a closed circle, the convex hull area increases and the calculated solidity decreases. Solidity is a dimensionless value.
Single cell metrics within Spheroids. Cell nuclei labeled with DAPI (Blue) provide total cell count for quantification of calcein-labeled live cells (Green) and PI-labeled (Red) dead cells.
Automated Spheroid analysis workflow. Tumorgenic cells (green) co-cultured with normal tissue-specific fibroblasts (Red).
